2-antiplasmin is an inhibitor of fibrinolysis, very fast binding plasmin, and irreversibly inhibiting
it. It is a single-stranded glycoprotein having a molecular weight of 60-70 kDt and migrating during electrophoresis 2-globulin. Principle. Method of the determination of
2-antiplasmin activity is based on its ability to neutralize of plasmin. Activity 2-antiplasmin plasma is determined by adding to her excess
plasmin’s. In this case, the inhibition of plasmin is proportional to the amount of 2-antiplasmin in the plasma. The remaining amount of
plasmin catalyzes the cleavage paranitroaniline (RNA) from a synthetic chromogenic substrate.
Absorption of free pNA determined at 405 nm is inversely proportional to activity 2-antiplasmin in
the plasma.
The process is as follows:
2-antiplasmin (plasma)+ plasmin (excess) ==>
( 2-antiplasmin-plasmin) + plasmin (residue).
Substrate – RNA + plasmin (residue) ==> Peptide
+ RNA
